报告简介:
Integrin-based focal adhesion complexes are composed of multiple focal adhesion proteins that form mechanical links between actin cytoskeleton and extracellular matrix. Embryonic stem cells (ESC) are unspecialized cells derived from blastocyst’s inner cell mass, which are able to differentiate into any cell type and possess the capability of self-renewal. Fibroblasts are ubiquitous cells of connective tissues that are thought to play critical roles in migration, and served as feeder cells to support ESC growth. Focal adhesion complexes and the underlying mechanosignaling are not well studied in both ESC and fibroblasts. In this study, integrative biochemical and biophysical approaches are adopted to compare the compositions and functions of focal adhesions in both cell types, with particular interest in one of the key focal adhesion proteins, Zyxin. Our preliminary findings highlight the critical role of zyxin as a mechanical sensor in fibroblast durotaxis and ESC differentiation.
